Corneal Confocal Microscopy: a Surrogate Endpoint for Neurodegeneration in Clinical Trials

Skin biopsy is currently considered the gold standard to define the diagnosis of small fibre neuropathy, even though results should be interpreted within the clinical context. Indeed, our recent validation study, while confirming the high specificity and sensitivity of skin biopsy, also demonstrated the reliability of combined clinical, functional and structural approach to the diagnosis of SFN. This lecture will discuss techniques and approaches to skin nerve quantification, impact of normative reference, and correlation with clinical features in order to provide reliable clues for clinical trial design.
